Blockade of IFNAR1 enhances IL-4 and IL-10 production by CD4+ T cells. NOD mice were treated i.p. with anti-IFNAR1 or control mAb on days 15 and 20 after birth, and killed at 25 days or 12 weeks of age (n = 4 per group). CD4+ T cells were purified from spleen and stimulated with anti-CD3 and CD28 mAbs for 48 h in vitro. (A) IL-4, IL-10, and IFN-γ secretion in culture supernatants by stimulated CD4+ T cells of 25-day-old mice was measured by ELISA. (B) Representative FACS plots show intracellular IL-10 staining in CD4+ T cells from anti-IFNAR1- or control mAb-treated 25-day-old NOD mice after stimulation with anti-CD3 and CD28 mAbs, followed by PMA and ionomycin.
