The amount and oligomerization state of Sll0822 protein in vivo. Ammonium-grown wild-type cells were harvested, washed, and inoculated into fresh medium of different nitrogen conditions. After 12 h, crude extracts were prepared from cells incubated in ammonium-containing medium (lane 1), nitrate-containing medium (lane 2), or nitrogen-free medium (lane 3) and treated with the cross-linker EDC. Total protein equivalent to 1 × 10⁷ cells per lane was separated by 15% (w/v) SDS-PAGE, and Sll0822 was detected by its specific antibody.
