Spc98p is phosphorylated after SPB duplication. Wild-type (CDC4) (A and B) and cdc4–1 cells (C and D) were synchronized with α-factor at 23°C and then shifted to 37°C in the absence of α-factor. Spc98p of cell lysates was enriched by immunoprecipitation. Spc98p phosphorylation (A and C) was analyzed by immunoblotting using anti-Spc98p antibodies. The DNA content (B and D) of the cells was analyzed over time by flow cytometry.
