Modular organization of (A) picromycin/methymycin synthase (PICS) and (B) 6-deoxyerythronolide B synthase (DEBS). In addition to the three core catalytic domains – the ketosynthase (KS), acyl transferase (AT), and acyl carrier protein (ACP) domains – individual modules carry specific combinations of auxiliary ketoreductase (KR), dehydratase (DH), and enoylreductase (ER) domains. Loading tri- or didomains prime module 1 of each synthase with the propionyl starter unit and dedicated thioesterase (TE) domains cyclize the mature polyketide with release of the corresponding macrolide aglycone.
