E1-G_TMCT and E1-G_CT are retained in a pre-Golgi compartment. Stably transfected CHO cells expressing E1, E1-G_CT, or E1-G_TMCT were pulse labeled for 15 min with ³⁵S-methionine/cysteine and then incubated for 0, 60, or 205 min in media containing excess methionine and cysteine. The cells were lysed and immunoprecipitated with human antirubella virus serum and protein A-Sepharose. The immune complexes were denatured, incubated at 37°C with or without endo H for 16 h, and then separated by SDS-PAGE on 10% gels followed by fluorography. Similar to E1, the bulk of E1-G_CT and E1-G_TMCT remains sensitive to endo H after 205 min.
