Ethanol pre-treatment (100 mM; 60 min) increased dopamine uptake velocity. Uptake of [3H]DA (10 nM-10μM) was measured after a 60 min exposure to either KRH (control; filled circles) or ethanol (100 mM; open circles) in DAT expressing HEK-293 cells. Vmax (control 78.2 pmol/min/well; ethanol 107.9 pmol/min/well) and Km (control 1.06 μM; ethanol 1.12 μM) parameter estimates were obtained from the nonlinear regression curve fits. The graph is representative of four experiments. Each data point represents an average of three wells ± SEM.
