In vivo effect of Sibelium™ on cisplatin-induced pro-inflammatory cytokine expression in the cochlea. Cochleas from Balb/c mice injected with PBS, cisplatin, or cisplatin plus Sibelum™ were removed and embedded in paraffin, then 5-μm sections were prepared. For immunohistochemistry studies, an immunohistochemistry kit (DAKO LSAB Universal K680, Carpinteria, CA, USA) was used, and all the procedures were carried out according to the manufacturer’s instructions. The following primary antibodies were employed: anti-TNF-α (goat polyclonal anti-mouse TNF-α, R&D Systems); anti-IL-1β (goat polyclonal anti-mouse IL-1β, R&D Systems); anti-IL-6 (goat polyclonal anti-mouse IL-6, R&D Systems); anti-NF-κB p65 (rabbit polyclonal IgG, Santa Cruz).
