Effects of pharmacological inhibition of MAPKs on cisplatin-induced secretion and mRNA expression of proinflammatory cytokines in HEI-OC1 auditory cells. Cells were treated with 20 μM cisplatin in the presence of 10 μM MAPK inhibitors as indicated below each plot. Proinflammatory cytokine secretion in culture supernatants was then determined at 1 and 24 h after cisplatin exposure by ELISA. A Release of TNF-α, B release of IL-1β, C release of IL-6. The data represent the mean ± SD of six culture supernatants. * and # p < 0.05 and ** and ## p < 0.01 compared with 1 h cisplatin exposure (* and **) or 24 h cisplatin exposure (# and ##). D Total RNA isolated from cells treated with cisplatin and MAPKs inhibitors for 24 h by TRIzol and cDNA was synthesized by reverse transcriptase. TNF-α, IL-1β, IL-6, and β-actin cDNA sequences were amplified using specific primer sets.
