Tyrosine kinase inhibitor genistein blocks KIM-127–stimulated polarization of lymphoblasts. (A) Lymphoblasts were washed in RPMI 1640 medium and then treated as follows: pretreated for 2 h with 0.3% DMSO and then plated on ICAM-1-Fc–coated dishes for 60 min (CONTROL); pretreated for 2 h with 0.3% DMSO, plated on ICAM-Fc–coated dishes, and stimulated for 60 min with 10 μg/ml mAb KIM-127 (KIM-127); and pretreated for 2 h with 30 μM of genistein dissolved in DMSO, plated on ICAM-Fc–coated dishes, and stimulated for 60 min with 10 μg/ml mAb KIM-127 (KIM-127 + GENISTEIN). Cells were fixed and stained with 0.5% crystal violet in 20% methanol before photography. A representative experiment is shown. (B) Quantification of the cell adhesion assays represented in A was carried out as specified in MATERIALS AND METHODS. The results are the mean ± SEM of three independent experiments.
