Quantitation of cell migration

T lymphoblasts were plated on ICAM-1-Fc–coated plastic dishes and allowed to adhere for 45 min in the presence of the indicated stimuli before filming starts. Films of cells were generated from stacks of individual digital images (frames) taken every 20 s for 2 h in an Zeiss Axiovert 135 video microscope using the IP-Lab-Spectrum software. The distances migrated by individual cells were quantitated using the Cell Tracking software extension developed by Tim Hutton. Numbers represent average speeds of migration of individual cells over a 20-min period in micrometers per hour.
