Summary of myosin II inhibition effects on myelination by SC and OL. Myosin II is an important regulator of SC cytoskeleton during differentiation and myelination. Myosin II appears to be necessary for the establishment of SC polarity and elongation along the axons, a crucial step in the initiation of myelin formation in the PNS. In the absence of myosin II activity, SC are unable to segregate axons in a 1:1 relationship and myelinate. In contrast, OL differentiation and myelin formation are independent of myosin II activity. Furthermore, down-regulation of myosin II promotes actin polymerization and branching, and also results in a significant increase in the amount of myelinated segments formed in culture.
