The N-terminal domain is dispensable for cell adhesion–dependent regulation. (A) Cells expressing wild-type FAK (lanes 1–3), dl 51–377 (lanes 4–6), or dl 1–361 (lanes 7–9) were analyzed. FAK was immunoprecipitated from lysates of subconfluent cells (lanes 1, 4, and 7) or cells plated on poly-l-lysine–coated plates (lanes 2, 5, and 8) or fibronectin-coated plates (lanes 3, 6, and 9) using BC4, and the immune complexes were analyzed as described in Figure 1. (B) Cells expressing wild-type FAK (lanes 1–3) or dl 721–857 (lanes 4–6) were analyzed. FAK was immunoprecipitated from lysates of cultured cells (lanes 1 and 4) or cells plated on poly-l-lysine–coated plates (lanes 2 and 5) or fibronectin-coated plates (lanes 3 and 6) using BC4. Samples were analyzed as described in Figure 1.
