Ascorbate and apocynin inhibit Jak2 and JNK activation. Endothelial cells were pretreated with ascorbate (Asc, 500 μM, 12 h) or apocynin (Apo, 250 μM, 1 h). Subsequently they were incubated with the same drugs plus LPS+IFNγ for another 30 min, and then were harvested for detection of total IκB protein levels and phosphorylation levels of Jak2, ERK and JNK by Western blot analysis. Total JNK (54 kDa JNK) served as loading control.
