Rad18p is a nuclear protein with an essential ATP-binding domain. (A) Rad18p localizes to the chromatin compartment of the nucleus. Cells expressing myc-tagged rad18p were stained with 9E10 anti-myc antibody, visualized with Cy3, and counterstained with DAPI. (B) rad18-DN is dominant negative when overexpressed. Wild-type colonies were grown in the presence (+T) or absence (−T) of thiamine for 3 d at 30°C. Plasmids: (1) pREP1 empty vector; (2) pREP1 wild-type rad18; (3) pREP1 rad18-DN. (C) rad18-DN cannot complement rad18-X. rad18-X cells were grown in the presence (+MMS) or absence (−MMS) of 0.0075% MMS on thiamine-containing media (which represses lethal expression) for 3 d at 30°C. Plasmids: (1) pREP1 empty vector; (2) pREP1 wild-type rad18; (3) pREP1 rad18-DN. rad18-DN cannot complement rad18-74 (our unpublished results).
