All strains not containing top2-191 or mis4-242 were grown to midlogarithmic phase at 30°C before fixation, DAPI staining, and observation by fluorescence microscopy. top2-191- and mis4-242-containing strains were grown to midlogarithmic phase at 25°C and then shifted to 30°C for 4 h before fixation, DAPI staining, and observation by fluorescence microscopy.
