MUC1 abrogates HGF-stimulated motility and invasion. S2-013.Neo, S2-013.MUC1F, or S2-013.MUC1.FHPM cells were added to the upper compartment of migration or invasion chamber in serum-free culture media. HGF-containing media were added to the lower chamber. After 24 h of incubation, cells in the upper chamber were removed, and cells that had migrated or invaded onto the lower surface of the membrane were stained and counted in 10 different fields of light microscopy. Bars represent mean ± S.E. of migrating (A) or invading cell number (B) from three independent experiments performed in triplicate (*** indicates p < 0.001 and ** indicates p < 0.01). To study the actin cytoskeleton changes, cells were serum-starved for 24 h or stimulated with HGF for 2 h post-serum starvation. The actin cytoskeleton was visualized with Alexa-488-conjugated phalloidin (green), and the nuclei were stained with propidium iodide (red) and analyzed by laser scanning confocal microscopy (A).
