Immunoblot and silver stain results for signaling proteins in HeLa cell lysate focused in a 3-10 pH gradient. Phospho-AKT and total ERK2 signal shows weaker focusing, with a maximum concentration factor of twofold. Cytochrome C is focuses strongly at a high pH, consistent with its high isoelectric point. As these proteins are focused, the total amount of protein, as determined by silver stain remains largely unchanged. These proteins are concentrated and collected from whole cell lysate at 83nL/s (18s of focusing) at ∼300V/cm.
