(A) Overexpression of cdc25⁺ rescues the cell elongation phenotype of Δcdr2. Δcdr2 (JK2240) cells were transformed with pREP3 (vector) or pREP3-cdc25⁺. The transformants were grown in EMM2 medium without thiamine at 30°C to induce the nmt1 promoter of the vector. Cells were photographed by differential interference contrast microscopy. Bar, 10 μm. (B) Growth of Δcdr2 cdc25–22 wee1–50 cells at restrictive temperature. Wild-type (PR109), cdc25–22 wee1–50 (KS1361), Δcdr2 (JK2240), Δcdr2 cdc25–22 wee1–50 (JK2243), Δslm9 (JK2246), and Δslm9 cdc25–22 wee1–50 (JK2248) cells were grown on YES media for 2–3 d at 25°C permissive temperature and 35°C restrictive temperature.
