A polyhistidine tag was added to the C terminus of the α-ACT variants. The α-ACT variants were purified from the conditioned medium by Ni-NTA column and examined by Coomassie blue and western blot analysis. (d and e) Inhibition by the α-ACT variants was assayed by incubation with proMMP-9 and skin extract, and the reactions were revealed by zymography.
