Cells (2× 104) were plated in 96 well plates in complete growth medium and treated with increasing concentration of 4-HNE (0-40μM) for 2h at 37°C. Same number of cells in duplicate columns of 96 well plates was separately treated for 2h with Fas-agonistic CH11 antibodies (50ng/well) and MTT cytotoxicity assay was performed as described in Materials and Methods section.
