The fluorescence intensity was monitored over an 8-h period for the GFP-mtX₁₂ variants with each of the last three tail positions modified individually or with the fourth and fifth positions modified together. The last 5 amino acids of the native DesA3 are DDLAA. The amino acid modified at each position is labeled as X. In order to analyze all these variants simultaneously in a time scale that would not be influenced by the observed degradation, a single experiment instead of triplicate experiments was performed. However, similar results were obtained in two other independent experiments (data not shown).
