Sequence characterization of cloned PCR products from direct TCRβ V-to-D rearrangements

PCR products shown in Figure 1A were cloned and sequenced. Two of the three possible Vβ8 gene segments (a single underlined G differentiates them) were detected in rearrangements with intact Dβ1 or Dβ2 gene segments. The third Vβ8 gene segment is not amplified by these primers. The RSS heptamer adjacent to each of the rearranged gene segments is shown (lower case). Non-templated nucleotides (N) and associated P nucleotides (P, underlined) are shown. Recovered sequences span several hundred basepairs on either side of the coding joints (data not shown). Table includes sequences of cloned VD PCR products from several other animals.
