Dtr1C15Δp-GFP is transported directly to vacuoles in vegetative cells. (A) Dtr1p-GFP was expressed under the control of the CLB2 promoter in wild-type (WT) (Y7479) and vps54Δ (Y7557) cells. Dtr1p-GFP in vps54Δ cells showed an identical GFP pattern, localized in the PM and vacuoles, to that in wild-type cells, suggesting that Dtr1p-GFP is not recycled through the endosome. (B) Dtr1C15Δp-GFP expressed in vegetative cells was detected at the vacuoles in wild-type cells (Y7777). An identical staining pattern was seen in the endocytosis-defective end3Δ mutant (Y7807), suggesting Dtr1C15Δp-GFP was not transported to the PM. Bars, 5 μm.
