Syntaxin 7 and syntaxin 8 form a complex that is regulated by NSF/α-SNAP. (A) HL-60 cell Triton X-100 extract was fractionated by velocity centrifugation through a 11–34% glycerol gradient. Fractions were separated by SDS-PAGE and analyzed by immunoblotting for the presence of syntaxin 7 (top panel) and syntaxin 8 (bottom panel). (B and C) HL-60 cell Triton X-100 extracts were incubated with (middle and bottom panels) or without (top panel) recombinant NSF and α-SNAP under conditions blocking (middle panel) or allowing (bottom panel) NSF ATPase activity. Extracts were then separated on 23–49% glycerol velocity gradients and blotted for syntaxin 7 (B) and syntaxin 8 (C).
