pH 3.4 was taken as 100% relative activity. B, effect of temperature
on TGP1 stability and activity. For stability experiments (broken
line), TGP1 was preincubated at various temperatures for 30 min, cooled
to 4 °C and assayed at 30 °C. Stability is measured relative to
samples stored on ice. For temperature optimum experiments (solid
line), end point fluorescence was monitored after 15 min of incubation of
enzyme and substrate at defined temperatures. Activity was calculated relative
to maximum fluorescence at 50 °C.
