FMRFa upregulates ap-uch mRNA and protein in pleural ganglia. A, QRT-PCR analysis of mRNA extracted from ganglia either immediately or 1 h after the end of 90 min treatment with FMRFa or vehicle (control). Immediately after the end of treatment, a significant increase in ap-uch mRNA was observed compared with time-matched control (*p < 0.05). In the presence of the p38 kinase inhibitor SB203580, FMRFa did not induce an increase in ap-uch mRNA. The effect of FMRFa on ap-uch was transient because 1 h after the end of treatment the mRNA was at control levels. B, Using an antibody specific for ap-uch, Western blot analysis was conducted on extracts from ganglia treated with FMRFa (FM) or vehicle [artificial seawater; control (C)] and lysed immediately, 1, 2, or 5 h after the end of treatment. Membranes were also probed with an anti-GAPDH antibody for normalization purposes. Compared with time-matched controls, ap-uch protein significantly increased only at the 1 h time point, although a tendency for increase was observed at all time points examined (*p < 0.05). These results indicate that the upregulation of ap-uch mRNA is accompanied by increased ap-uch protein synthesis. Error bars indicate SEM.
