H77 NS4B does not support Con1 replication. (A) Huh-7.5 cells were electroporated with 1 μg of replicon RNA and at 24, 48, 72, and 96 h posttransfection total cellular RNA was harvested. HCV RNA was quantified by real-time RT-PCR and the number of HCV RNA molecules was normalized to 1 μg of total cellular RNA (HCV RNA/μg total cell RNA). (B) At 96 h postelectroporation, Huh-7.5 cell monolayers were lysed and the relative levels of NS5A expression were visualized by Western blotting. The migration of NS5A is shown on the left.
