Analysis of the replication and damage checkpoint of rfc3-1 cells at permissive temperature (26°C). Synchronous cultures of wild-type (h− L972) and rfc3-1 (MSY1) cells were generated by elutriation and incubated in the presence of 10 mM HU (A) and 0.01% MMS (B) or exposed to gamma (C) and UV (D) irradiation. The percentages of septated cells or cells passing mitosis were estimated by DAPI and calcofluor staining.
