H460 tumor cells were cultured in a 6-well plate (2×10⁶/well/2 ml) in the presence or absence of doxorubicin (0.125 μg/ml) for 24h. Supernatants were collected, remaining cells were harvested and sonicated, and tumor lysates were centrifuged to remove cell debris. Tumor cell lysates (equivalent to 1×10⁶tumor cells) were added to 1 ml of fresh culture medium. The concentrations of cytokines in media conditioned by untreated tumor cells (TCM), doxorubicin-treated tumor cells (DTCM) or fresh media (FM) conditioned by tumor lysates were determined using a Luminex multiplex assay.
