Effect of selected arylcarboxylic acid hydrazides on TNF-α production by murine J774.A1 macrophages. Panel A: Macrophages (2×105 cells/well) were cultured in the presence buffer (0) or the indicated concentrations of Compound 1 (●), 2 (○), 3 (■), 4 (□), 5 (◆), AG-14 (△), or LPS (2 ng/ml) (◇) for 24 hr, and TNF-α was measured in the cell supernatants by ELISA. Panel B: Macrophages (2×105 cell/well) were cultured in the presence of the 12.5 µM Compound 1 (●), 2 (○), of DMSO (■) for the indicated times, and TNF-α was measured in the cell supernatants by ELISA. The data in both panels are presented as the mean±SD of triplicate samples from one experiment, which is representative of three independent experiments.
