Cyclin G2 inhibits cell proliferation. (A) Measurement of cell proliferation by BrdU assay. OV2008 cells were transiently transfected with CCNG2-V5 plasmid. Overexpression of CCNG2 resulted in a decrease in cell proliferation in a dose- and time-dependent manner. (B) Analysis of cell cycle profile by flow cytometry. OV2008 cells were transfected with an empty vector (EV), ALK7-ca, or CCNG2-V5. Both ALK7-ca and cyclin G2 increased the number of cells in G1 phase and decreased the number of cells in S phase. (C) Transfection efficiency. OV2008 cells were cotransfected with pEGFP and an empty vector (EV), ALK7-ca, or CCNG2-V5 constructs. More than 60% of GFP-positive cells were observed after transfection with these plasmids. At 2 d after transfection, cell density was lower in cyclin G2- and ALK7-ca–transfected cells.
