Loss of [URE3] upon Sis1 depletion. (A) sis1-Δ [TETrSIS1] [URE3] carrying either an extra copy of SIS1 under control of its own promoter in a low copy plasmid (squares) or an empty vector (diamonds). Cells were harvested after the indicated number of generations in the presence of doxycycline and plated onto rich media. Left: The percentage of [URE3] (white) versus [ure-o] (red) colonies was determined and plotted. The dotted line represents kinetic modeling of 25 seeds/cell and no fragmentation. Right: Representative sample of sis1-Δ (TETrSIS1) [URE3] cells, which have an additional copy of SIS1 or an empty vector (−), taken at 12 and 10 generations, respectively.
