NEURON simulations reveal how individual kinetic changes to A currents caused by DPPX contribute to specific firing properties of hippocampal CA1 neurons. From the control simulation to the DPPX siRNA simulation, onset of the 1st AP increased from 17.9 to 30.2 ms. In addition, the peak amplitude of the 1st AP decreased from 108.4 to 107.2 mV, the 1st AP afterhyperpolarization potential decreased from −1.4 to −0.4 mV, and the 1st AP half-width increased from 1.9 to 2.0 ms. No change in frequency was found after the 1st few APs. These findings are all consistent with experimental results. Traces in A are expanded in B.
