HRASLS3 overexpression increases lipid accumulation and adipocyte-associated gene expression. Stable cell lines ectopically expressing HRASLS3 undergo enhanced adipogenesis as demonstrated by increased lipid accumulation (A) and higher expression of a number of adipogenic markers (B). NIH-3T3 cells stably expressing PPARγ were infected with retrovirus containing a pBabe HRASL3 expression vector to create cell lines stable expressing HRASLS3 (B). Cells were grown to confluence and induced to differentiate by treatment with dexamethasone (1 μM) for 48 h and continuous treatment with the synthetic PPARγ ligand GW7845 (100 nM). At the time points indicated, cells were fixed and stained with Oil Red O (A). In addition, 6 days after induction of differentiation mRNA was isolated and expression of adipogenic markers examined by real-time quantitative PCR (B).
