GnRHa-Induced Pyk2 Phosphorylation Is Blocked by Cam Inhibition

αT3-1 cells were pretreated with saline (control) or the Cam inhibitor W7 (15 μm) for 30 min. Cells then received GnRHa for 15 or 30 min, and whole-cell lysates were prepared. Western blot analysis was used to determine the phosphorylation status of Pyk2 (p-Y⁴⁰²). A, The Pan-Pyk2 antibody was used to determine lane loading. B, A similar study was performed except cells were pretreated with increasing doses of the CamK II-specific inhibitor KN62 (5 or 10 μm), and Western blots were used to determine phosphorylation status of Pyk2 as described above.
