EBV BGLF4 kinase interacts with lamin A/C and induces the disassembly of the nuclear lamina accompanied by chromosome condensation. (A) HeLa cells were transfected with plasmid expressing BGLF4, K102I, or a control vector. At 24 h posttransfection, cells were fixed with 4% paraformaldehyde and stained for BGLF4 (red), lamin A/C (green), and DNA (blue) as described in Materials and Methods. BGLF4-expressing cells showed disrupted nuclear lamina with intranuclear aggregates of lamin A/C accompanied by condensed chromosomes. BGLF4 colocalized with lamin A/C in the nucleus (yellow in the merge images). (B) Lysates from HeLa cells with BGLF4 or K102I expression were immunoprecipitated with antibody against lamin A/C (α-lamin A/C) or GST (α-GST).
