The Ca2+ rise elicited by 100 μM H2O2 is attenuated in cells expressing P1018L. Ca2+ measurements were performed on Fura-2 loaded HEK-293 cells induced to express WT (black) and P1018L (gray), and in uninduced cells (broken lines) at RT (A) and 37°C (B). The magnitude of the Ca2+ rise in WT was ≈1.2 times greater at 37°C than at RT, and 1.1 times greater at 37 °C than RT for P1018L. The experiments shown in A and B were done on the same day. (C) H2O2 (100 μM) did not elicit a similar Ca2+ rise in untransfected parental cells, TR-293, which are HEK-293 cells stably expressing the tetracycline repressor (Invitrogen) (D) No measurable Ca2+ response to 100 μM H2O2 is seen in the presence of EGTA.
