Oligonucleotide masking of Gtx-binding site in 18S rRNA blocks activity of recombinant mRNAs. Plasmids containing Gtx (pGtx)(A) or FGF2 (pFGF2)(B)5′ leader and coding sequences were transiently transfected into COS-7 cells and cell lysates analyzed by Western blot analysis using anti-Gtx, anti-FGF2, or anti-β-actin antibodies as indicated. Histogram above Western blots indicates plasmids and oligonucleotides (oligo) in the various samples. Molecular weights of Gtx, FGF2, and β-actin protein bands are indicated to the right. This experiment was repeated with similar results three times.
