Separase stabilises microtubule dynamics at anaphase onset. A photobleached segment (yellow box) of the GFP-tubulin labelled spindle recovers in strain Y1363 (MATa MET-CDC20 GAL-ESP1 yEGFP-TUB1) arrested in metaphase, indicative of high microtubule turnover, but recovery is reduced after anaphase onset triggered by separase. After TEV protease-triggered anaphase in strain Y1362 (MATα MET-CDC20 GAL-TEV SCC1TEV yEGFP-TUB1) fluorescence recovery remains high. Fluorescence intensity in control (white box, open circles) and bleached (red box, filled circles) regions are plotted relative to their pre-bleach intensity.
