Selected characteristics of HC11-C4 and HC11-C24 cells are shown. *, spontaneous cell death rate was determined after growing cells in standard rich medium containing 10% FCS and then shifting the cells into serum free medium (SFM). Cells were grown for 48hrs following the shift into SFM and spontaneous cell death was determined following trypsinization and washing using Trypan Blue exclusion and manual counting with a hemocytometer. #, doubling time for each cell line was determined by plating at a density of 3.0 × 105 cells per well in six well trays. Trypsinized cells were counted manually 24 and 48 hours later using a hemocytometer. Response to TGFβ was determined by western immunoblot (α-SMA, E-cadherin) as shown above or by visual inspection of photomicrographs (phalloid stain for F-actin fiber formation). XOR activity was determined as shown in Fig. 4F.
