Overexpression of DsbA-L increases cellular adiponectin levels and expression. 3T3- L1 CAR adipocytes were infected with adenoviruses encoding GFP or GFP plus DsbA-L. Thirty-six hours after infection, cells were lysed and proteins in cell lysates were separated by SDS/PAGE and determined by Western blot by using specific antibodies as indicated (A). Adiponectin in cell lysates (B) or cell culture medium (D) were resolved by gel-filtration chromatography and determined by Western blot by using an antibody to adiponectin. Data are representatives of 3 independent experiments with similar results. Quantification of the relative abundance of adiponectin oligomers in cell lysates (C) and culture medium (E) was performed by analyzing Western blots by using the National Institutes of Health Scion Image software.
