Insulin responsive site of HO-1 promoter is located downstream of bp -2986. BRPC were transiently transfected with deletion mutants of the HO-1 promoter (250 ng/well). After the transfection, the cells were treated with insulin (A, 100 nm) or H₂O₂ (B, 50 μm) for 24 h and then harvested. HO-1 promoter activity was calculated by comparison with the HO-1-(-4045/+74) promoter construct. ^*, p < 0.05 versus activity of HO-1-(-4045/+74) promoter construct stimulated with either insulin or H₂O₂ treatment in three separate experiments.
