Reduction of NF-κB binding activity induced by
H2O2 by insulin through IRS1. BRPC were seeded in a
6-well culture plate, transfected with control GFP, wild-type IRS1, or IRS2,
incubated in the absence or presence of insulin (100 nm) for 12 h,
and then exposed to H2O2 for 1 h as described under
“Experimental Procedures.” NF-κB binding activity assay was
performed according to the manufacturer's instructions as described under
“Experimental Procedures.” The results are shown as the means
± S.D. of three independent experiments. *, p <
0.001 versus PBS-treated cells; †, p < 0.01
versus H2O2.
