L1 and NL are each necessary for maintaining innervation of synaptic α3*-nAChRs on transfected neurons. Electroporated E14 CG neurons were coimmunostained for SV2 and α3*-nAChRs, followed by quantitative image analysis as in Fig. 4. Neurons transfected with L1Cyt or NL54 had significantly lower total numbers of α3*-nAChR clusters overlaid by SV2 puncta (A), apparent also as a reduced fraction of α3*-nAChR clusters overlaid by SV2 puncta (B). SynCyt did not significantly reduce either. Neither SynCyt nor LiCyt changed either the total number or the proportion of α7-nAChR clusters overlaid by SV2 puncta (C). *p < 0.05; **p < 0.01 compared to GFP by ANOVA with Bonferroni post-tests. Number of cells analyzed were as in Fig. 4 for panels A and B; for panel C, the number was 8, 27, and 24 for GFP, L1Cyt, and SynCyt, respectively (1–4 cells per ganglion), plus 2–4 unelectroporated control cells in the same section for each electroporated cell.
