CCK regulates Y2R expression in luciferase promoter-reporter assays. A, In cultured vagal afferent neurons transfected with empty vector (pGL4), 10 nm CCK8s for 16 h had no effect on luciferase activity, whereas there was significant stimulation of luciferase activity in neurons transfected with Y2R-luc (Y2Rpr). B, In vagal afferent neurons transfected with Y2R-luc, the response to 10 nm CCK8s was inhibited by 1 μm Ro-32,0432. C, PMA (100 nm) also stimulated Y2R expression, and this was inhibited by Ro-32,0432. D, Ghrelin inhibited CCK8s stimulation of Y2R-luc expression in cultured vagal afferent neurons.
