Reduction of ISP induced by stigmatellin in the absence of exogenous electron donor. The bc1 complexes from the wild type, S141C(ISP)/G180C(cyt c1), and A185C(cyt b)/K70C(ISP) were diluted to 10 μM in 3 ml of 20 mM Tris-Cl buffer, pH 8.0, containing 200 mM NaCl and 0.01% LM. After incubated with 50 μM stigmatellin for 5 min, 1 hr, and 2 hr, the bc1 complexes was subjected to the CD to measure the reduction of ISP.
