In each temperature-sensitive mutant strain identified in this study, a Km resistance gene was inserted near the ltaS gene locus, and the resultant mutant was used as a donor for transduction analysis using phage 80alpha. The recipient was M0107 in all transductions. The Km-resistant (Km^r) transductants were selected at 30°C and tested for temperature-sensitive cell growth (TS phenotype) at 43°C. Data are representative of two independent experiments.
