Effect of paraformaldehyde fixed parasites (Ld-F) or freeze-thawed parasites (Ld-K) infection on Pam3cys stimulated activation of MAPK pathway by THP-1 cells. THP-1 cells were infected with either Ld-F or Ld-K simultaneously activated with Pam3cys (1ug/ml) for 0, 5 and 15 min. Cells lysates were subjected to electrophoresis, transferred on nitrocellulose membrane and probed with antibodies to phosphorylated and non-phosphorylated P³⁸ and ERK1/2. Figure shown is a representative of three independent experiments (A). Data shows ratio of densitometric reading phosphorylated P³⁸and ERK1/2 to non-phosphorylated P³⁸ (B) and ERK1/2 (C). Mean + SD of three independent experiments is shown and group comparison was done using student's t test P < 0·05 considered significant (*P < 0·05, ***P < 0·005).
