Rapid localization of siRNA to P-bodies. (A) HeLa cells grown on poly-l-lysine–coated coverslips were transfected with 50 nM Cy3 siPPIB, Cy3 siSMN, or Cy3 siLuc (Red). At the given time points after transfection, cells were fixed and immunostained with anti-GWB serum (green). (B) Colocalization of Cy3 siPPIB to P-bodies was statistically quantified using the Jacop program from six images, three containing between three and five cells per image and three containing ∼10 cells per image. Significant colocalization was observed, with the average Pearson's colocalization coefficient, r = 0.268 (SE = 0.038), p value for statistically significant colocalization obtained with Costes randomization based colocalization for each individual image <0.01.
