The pmrA constitutive parental strain MST100 (1), as well as AY100 (ΔpmrL) (2), AY101(ΔpmrM)(3), and AY103 (ΔarnT)(4) were grown to mid-log phase. The washed cells were treated with NHS-biotin at 2 mM for 4 h at 4°C. Reactions were quenched with 50 mM glycine. Lipids were extracted and subjected to mild alkaline hydrolysis (50). The hydrolysis products were separated by TLC, using the solvent chloroform/methanol/acetic acid/H2O (25:15:4:2, v/v), and visualized by charring as in Fig. 5.
